Two- and three-dimensional transient melt-flow simulation in vapour-pressure-controlled Czochralski crystal growth

Flow and thermal properties associated with semiconductor melt flow in an axisymmetric crucible container are studied numerically. Axisymmetric and three-dimensional computational solutions are obtained using a standard-Galerkin, finite-element solver. The crucible and crystal are optionally rotated, and the influence of gravity (through buoyancy) is accounted for via a Boussinesq approximation in the controlling Navier-Stokes equations. The results indicate a strong dependence of the flow on both rotation and buoyancy. Results for axisymmetric flows, computed in both flat and curved geometries, are presented first, and strongly suggest that rotation of crystal and crucible in the same direction (iso-rotation) is most favourable for producing a desired convexity for the crystal/melt interface. Three-dimensional results are then presented for higher Reynolds numbers, and, in particular, reveal that for iso-rotation under moderate buoyancy, the flow undergoes a switch from a steady, 2D state to an unsteady 3D state, and that the temperature becomes non-trivially advected by the flow beneath the crystal. Further evidence reveals however, that on a time scale more appropriate to the crystal growth process, the (time-averaged) flow has a weaker three-dimensionality, in relation to its axisymmetric mode, and there is only slight distortion to the temperature field beneath the crystal

Characterisation of chitin in the cuticle of a velvet worm (Onychophora)

*Kaya, Murat ( Aksaray, Yazar ) *Baran, Talat ( Aksaray, Yazar )We characterize the trunk cuticle of velvet worms of the Peripatoides novaezealandiae-group (Onychophora) using SEM, TEM, Fourier transform infrared spectroscopy (FT-IR), and thermogravimetric analysis (TGA). TEM and SEM revealed a relatively uniform organization of the delicate cuticle that is covered by numerous bristled and nonbristled papillae with ribbed scales arranged in transverse rows. The cuticle consists of a very thin multilayered epicuticle of varying appearance followed by the largely fibrous procuticle. The irregularly arranged nanofibres of isolated cuticular chitin seen by SEM are considered as bundles of chitin fibres. FT-IR and TGA showed that the chitin is of the α-type. This confirms and broadens the single previous study in which the presence of α-chitin in a velvet worm was demonstrated with a single analysis (X-ray diffraction)...

    Get PDF

Endogenous tumor suppressor microRNA-193b: Therapeutic and prognostic value in acute myeloid leukemia

Purpose Dysregulated microRNAs are implicated in the pathogenesis and aggressiveness of acute myeloid leukemia (AML). We describe the effect of the hematopoietic stem-cell self-renewal regulating miR-193b on progression and prognosis of AML. Methods We profiled miR-193b-5p/3p expression in cytogenetically and clinically characterized de novo pediatric AML (n = 161) via quantitative real-time polymerase chain reaction and validated our findings in an independent cohort of 187 adult patients. We investigated the tumor suppressive function of miR-193b in human AML blasts, patient-derived xenografts, and miR-193b knockout mice in vitro and in vivo. Results miR-193b exerted important, endogenous, tumor-suppressive functions on the hematopoietic system. miR-193b-3p was downregulated in several cytogenetically defined subgroups of pediatric and adult AML, and low expression served as an independent indicator for poor prognosis in pediatric AML (risk ratio 6 standard error, 20.56 6 0.23; P = .016). miR-193b-3p expression improved the prognostic value of the European LeukemiaNet risk-group stratification or a 17-gene leukemic stemness score. In knockout mice, loss of miR-193b cooperated with Hoxa9/Meis1 during leukemogenesis, whereas restoring miR-193b expression impaired leukemic engraftment. Similarly, expression of miR-193b in AML blasts from patients diminished leukemic growth in vitro and in mouse xenografts. Mechanistically, miR-193b induced apoptosis and a G1/S-phase block in various human AML subgroups by targeting multiple factors of the KIT-RAS-RAF-MEK-ERK (MAPK) signaling cascade and the downstream cell cycle regulator CCND1. Conclusion The tumor-suppressive function is independent of patient age or genetics; therefore, restoring miR-193b would assure high antileukemic efficacy by blocking the entire MAPK signaling cascade while preventing the emergence of resistance mechanisms
